Lifespan
Longevity Benefits of Rapamycin
Rapamycin stands out as a pioneering medication that inhibits the formation of senescent cells—aged, dysfunctional cells that accumulate over time—potentially slowing aging and enhancing cellular performance across your body. While it has been an FDA-approved drug for a long time, many are now using it off-label for longevity purposes. Here are some reasons why:
Stimulate Autophagy: Activate your cells' self-cleaning mechanism, promoting the removal of dysfunctional cellular components and enhancing organelle turnover.
Tamp Down Senescence: Selectively inhibit mTORC1, a key component of the mTOR pathway, to mitigate cellular senescence and potentially slow age-related tissue dysfunction.
Reduce Inflammation: Decrease senescence-associated secretory phenotype (SASP), potentially alleviating chronic low-grade inflammation characteristic of aging.
Enhance the Immune System: Modulate immunosenescence, potentially improving T-cell function and bolstering your body's adaptive immune responses.
Improve Cognitive Performance: Support neuroplasticity and synaptic function, potentially enhancing memory consolidation and cognitive resilience.

Does Lifespan test medications for purity and bioavailability?
At Lifespan, we are committed to ensuring the effectiveness of the medications we prescribe. This commitment extends to addressing the challenges of bioavailability, which can notably impact the usage and efficiency of certain drugs, including rapamycin. To tackle this, we employ a robust testing program to select the formulations and brands of rapamycin that offer the best absorption levels. We believe in personalized care and thus, offer our patients the option to test their individual rapamycin levels. This enables us to understand each patient's unique absorption profile of the medication, taking into account the variability of their biology. This approach ensures that our patients receive the most effective therapeutic regimen tailored to their individual needs.

This personalized approach allows us to provide you with tailored information and insights to support your well-being effectively.
How frequently do I take my rapamycin dose on the longevity protocol?
Upon thorough medical review and screening, should you be deemed suitable for the Lifespan rapamycin protocol, your administration of Rapamycin will occur just once a week. For instance, should your current weekly dosage be 5mg, and you're given 1mg pills, you'll consume all five pills on a single day, as opposed to distributing them over the week. You'll then rest until the next week. The reason for this methodology stems from the cyclical nature of the benefits rapamycin offers for anti-aging. Unlike transplant patients who take rapamycin daily, the longevity benefits are drawn from the intermittent inhibition of mTOR. The therapeutic benefit of rapamycin is derived from pulsated inhibition of mTOR, avoiding continuous inhibition. This approach prevents bloodstream concentrations of rapamycin that could inhibit mTOR-Complex 2, leading to immune suppression, which we aim to evade.By adhering to a weekly dosage, adjusted according to your unique biomarkers and phenotype, we sidestep the immunosuppressive side effects of rapamycin. We strive to administer rapamycin in a manner that leverages the mTOR inhibitory benefits, such as stimulating autophagy, in cycles, and then returning to baseline mTOR activity.
What is the dose of the Lifespan rapamycin protocol?
At Lifespan, we harness the power of advanced technology and your unique biomarkers to craft a tailored rapamycin dosage plan just for you. Typically, we will initiate a patient's treatment with a weekly dose of 2-3mg, carefully escalating this over time to reach the optimum dosage, which generally lies between 5 to 8mg per week. Please note, the provision of medication is strictly contingent upon a comprehensive medical review and initial screening process.
Why do some sources mention a risk of cancer associated with rapamycin?
The risk of cancer is related to daily dosing for transplant patients, whereas our approach involves intermittent dosing every 7 days to avoid immune suppression. This distinction has varying implications for cancer. Rapamycin inhibits mTOR, exhibiting anti-cancer properties by preventing cell proliferation and inflammatory conditions conducive to cancer. There are two complexes of the mTOR pathway: mTORC1 and mTORC2. Daily dosing may suppress the immune system, while our low-dose cyclical dosing enhances immune function, potentially contributing to rapamycin's anti-cancer benefits. Inhibiting mTORC1, as low-dose cyclical dosing rapamycin does, has been shown to extend lifespan in various animal models by promoting autophagy, reducing inflammation, and slowing the aging process. Consistent, daily use of rapamycin will also inhibit mTORC2 and can bring on unwanted side effects. Our low-dose, cyclical dosing strategy harnesses the longevity-promoting effects of mTORC1 while avoiding the negative side effects linked to mTORC2 suppression.
